چکیده

3-Ketovalidoxylamine A C-N lyase and glucoside 3-dehydrogenase (G3DH), two key enzymes for valienamine synthesis, are produced by Stenotrophomonas maltrophilia. The condition of producing 3ketovalidoxylamine A C-N lyase and G3DH was optimized. Validamycin A was showed to be suitable carbon source for C-N lyase and G3DH synthesis. Maximum C-N lyase production was achieved by adding 100 ml culture medium into a 500 ml conical flask containing 5 mg/ml validamycin A, with 12% inoculation seed volume for 22 h with a productivity of 6.26 × 10 U/mg protein. The specific G3DH activity was 0.244 U/mg protein in the same culture condition at 27 h. A 15 L fermenter containing 8 liter of medium was also used in this study. Crude enzyme reaction showed the conversion of N-pnitrophenylvalidamine was achieved to 92.8% at 3 h and the yield of p-nitroaniline was 81.3% at 3 h.

Biochemical and computational study of an alginate lyase produced by Pseudomonas aeruginosa strain S21

Objective(s): Alginates play a key role in mucoid Pseudomonas aeruginosa colonization, biofilm formation, and driving out of cationic antibiotics. P. aeruginosa alginate lyase (AlgL) is a periplasmic enzyme that is necessary for alginate synthesis and secretion. It also has a role in depolymerization of alginates. Using AlgLs in cystic fibrosis patients along with anti...

Screening of Alginate Lyase-Producing Bacteria and Optimization of Media Compositions for Extracellular Alginate Lyase Production

Background: Alginate is a linear polysaccharide consisting of guluronate (polyG) and mannuronate (polyM) subunits. Methods: In the initial screening of alginate-degrading bacteria from soil, 10 isolates were able to grow on minimal medium containing alginate. The optimization of cell growth and alginate lyase (algL) production was carried out by the addition of 0.8% alginate and 0.2-0.3 M NaCl ...

Production of Recombinant Proline Dehydrogenase Enzyme from Pseudomonas fluorescens pf-5 in E. coli System

Proline dehydrogenase (ProDH; 1.5.99.8) belongs to superfamily of amino acid dehydrogenase, which plays a significant role in the metabolic pathway from proline to glutamate. The goal of this research was gene cloning and characterization of ProDH enzyme from Pseudomonas fluorescens pf-5 strain. The gene encoding ProDH was isolated by means of PCR amplification and cloned in an IPTG inducible T...
